This is actually the preferred route to Duck Hole from Upper Works. The trail should be well marked, though I can't confirm it, as I have never hiked that trail.

Hunting is permitted in the area, although the only season that will be open this weekend in that area is early bear.
 
Trail used to be very well marked. You will need to hop across Roaring Brook just before you get to DH. It might be a little confusing at the start, until you get to the Indian Pass cutoff, as there used to be some logging in there, but otherwise, it is a pretty straight forward trail.

I paddled/carried a canoe in there earlier this summer via Henderson and Preston. The trail sections between waterways were very well traveled and marked. I can only assume the trails around the lakes are equally obvious.

I personally wouldn't worry about hunters back in there at all, especially being before regular season opens. Wearing orange however never hurts at this time of year.

I have hiked it from DH to Henderson as a part of an almost-loop, but not in the reverse. It is the shortest route to DH. My recollection is that it was a very straightforward trail, and no problem with route finding. When markers are few, the trail is obvious. There was some beaver activity shortly after DH (heading toward Henderson), but nothing too outrageous. There are warnings about the difficulty of crossing the river in times of high water, but it was not an issue for us then, and I'm guessing it's not an issue now.
 
I've hiked it frequently. Generally well marked and easy to follow. Make sure to take the left and cross the bridge early on so you don't go to Indian Pass. A couple of the "Private Trail" junctions are confusingly signed, but the right way is pretty obvious.
